HEC201625 is an orally bioavailable small molecule inhibitor of programmed death-ligand 1 (PD-L1), developed by Sunshine Lake Pharma Co., Ltd. for the treatment of various tumors. Unlike monoclonal antibodies that block the PD-1/PD-L1 interaction extracellularly, HEC201625 binds specifically to PD-L1 on the surface of tumor cells, inducing its dimerization and subsequent internalization. This mechanism effectively prevents PD-L1 from interacting with the PD-1 receptor on immune T cells, thereby restoring the immune system's ability to recognize and eliminate tumor cells. Preclinical data have demonstrated that HEC201625 exhibits potent antitumor activity comparable or superior to established PD-L1 antibodies across multiple models, with the added benefits of oral administration and potentially improved tumor tissue penetration.
