HEC81885 PTSA is an orally bioavailable small molecule inhibitor of the Ataxia Telangiectasia and Rad3-related (ATR) kinase, developed by Guangdong Dongyangguang Pharmaceutical (HEC Pharm). ATR is a critical component of the DNA damage response (DDR) system, primarily responding to replication stress and single-strand DNA breaks. By inhibiting ATR, HEC81885 disrupts cell cycle checkpoint signaling and DNA repair processes, leading to the accumulation of DNA damage and subsequent apoptosis, particularly in tumor cells with high replication stress or deficiencies in other DDR components like ATM or p53. The drug is currently in Phase 1 clinical development for patients with advanced solid tumors to evaluate its safety, pharmacokinetics, and preliminary efficacy.
