HEC88473 is a novel long-acting dual agonist fusion protein targeting both the glucagon-like peptide 1 receptor (GLP-1R) and fibroblast growth factor 21 receptor (FGF21R). It is an Fc fusion protein containing FGF21 and GLP-1 moieties fused to the N-terminal and C-terminal of the Fc fragment, respectively. The drug is designed for subcutaneous injection and aims to provide comprehensive metabolic benefits by combining glucose-lowering effects with improvements in lipid metabolism and liver fat reduction. Clinical studies have shown that HEC88473 reduces hepatic fat fraction, improves glycemic control, insulin resistance, lipid profiles, and increases adiponectin levels. It has demonstrated good safety and tolerability in healthy subjects as well as patients with type 2 diabetes mellitus (T2DM), obesity, and metabolic dysfunction-associated steatotic liver disease (MASLD)/nonalcoholic steatohepatitis (NASH)[1][2][6][7].
