HEC95468 is a **novel, oral small molecule soluble guanylate cyclase (sGC) stimulator** in clinical development as a potential best-in-class therapy for the treatment of heart failure (HF) and pulmonary arterial hypertension (PAH). The drug enhances sGC activity independently of nitric oxide, thereby increasing cyclic guanosine monophosphate (cGMP) levels, leading to vasodilation, reduced blood pressure, and attenuation of cardiac and renal damage. It has demonstrated dose-proportional pharmacokinetics, increases cGMP concentrations, decreases systolic and diastolic blood pressure, and elevates compensatory vasoactive hormones. Preclinical data also show organ-protective effects in models of cardiorenal disorder, comparable or superior to established sGC stimulators such as riociguat and vericiguat.
