Overview

HEC96719 is a novel tricyclic small molecule that acts as a potent and selective agonist of the farnesoid X receptor (FXR), a nuclear bile acid receptor. It was developed to address non-alcoholic steatohepatitis (NASH), a progressive form of non-alcoholic fatty liver disease (NAFLD) that can lead to cirrhosis and hepatocellular carcinoma. Preclinical studies demonstrated that HEC96719 has superior potency compared to GW4064 and obeticholic acid in activating FXR, with higher selectivity and favorable tissue distribution predominantly in the liver and intestine. The drug has advanced through phase I safety trials in Australia and China, with phase II clinical trials completed for NASH. Sunshine Lake Pharma, part of the HEC Pharm Group, is responsible for its development[1][2][3][4][5][7].
