Overview

hEGFRvIII-CD3 (BRiTE) is a bispecific T-cell engager (BiTE) designed for the treatment of EGFRvIII-positive glioblastoma. It is a bi-scFv construct that simultaneously targets the CD3 epsilon subunit on T cells and the tumor-specific epidermal growth factor receptor variant III (EGFRvIII) epitope. By crosslinking T cells with EGFRvIII-expressing tumor cells, BRiTE redirects the patient's T-cell repertoire to specifically lyse glioblastoma cells in a TCR-agnostic manner. Developed under the leadership of Dr. Mustafa Khasraw at Duke University, the drug is an off-the-shelf immunotherapy administered via continuous intravenous infusion to overcome its short half-life. It is currently being evaluated in a Phase 1 clinical trial (NCT04903795) as monotherapy or in combination with autologous T-cell infusion for patients with newly diagnosed or recurrent glioblastoma.
