Overview

HEK5-1B3 is a monoclonal antibody clone that targets B7-H3 (CD276), an immune checkpoint molecule frequently overexpressed in acute myeloid leukemia (AML) and various solid tumors. Developed through research at the University of Texas MD Anderson Cancer Center and the University of Tübingen, HEK5-1B3 functions by blocking the immunomodulatory activity of B7-H3, which typically serves to inhibit immune cell responses. By neutralizing B7-H3, the antibody enhances natural killer (NK) cell-mediated cytotoxicity against AML cells. Preclinical evaluations in AML cell lines and patient-derived xenograft (PDX) models have shown that HEK5-1B3 can inhibit tumor growth and improve survival outcomes, although it was noted to be less potent than the T-1A5 clone in certain in vivo models.
