Helicore GIP-GLP-1 antibody conjugate

Helicore Biopharma is developing a portfolio of GIP-targeting antibody conjugates for the treatment of obesity and related metabolic conditions. This specific candidate is an antibody-drug conjugate (ADC) that combines a monoclonal antibody targeting the GIP ligand (potentially based on the company's HCR-188 lead) with a GLP-1 receptor agonist payload. The mechanism of action involves dual modulation of metabolic pathways: neutralizing the GIP ligand to improve insulin sensitivity and lipid metabolism, while simultaneously activating the GLP-1 receptor to suppress appetite and promote weight loss. This approach aims to provide superior efficacy and a more favorable dosing profile compared to existing peptide-based co-agonists.
