Hematopoietic stem and progenitor cell-derived natural killer cells are **natural killer (NK) cells generated in vitro from hematopoietic stem cells (HSCs) or progenitor cells** for adoptive immunotherapy. These NK cells are produced from CD34+ hematopoietic progenitors sourced from bone marrow, peripheral blood, umbilical cord blood, or pluripotent stem cells. Their mechanism of action involves innate immune surveillance through recognition and cytolytic elimination of virally infected or tumor cells, employing activating and inhibitory receptors, cytokine secretion (e.g., interferon gamma), cell lysis via perforin/granzyme, and death receptor pathways (TRAIL, FasL). HSPC-derived NK cells may offer a homogeneous, potentially expandable, and genetically modifiable source for cancer immunotherapy, overcoming some of the limitations of donor heterogeneity and limited cell numbers available from peripheral blood or umbilical cord blood NK cells. They are being researched as cellular immunotherapy for hematologic malignancies and solid tumors, often in the post-transplant setting or as a universal “off-the-shelf” allogeneic therapy[3][4].
