Overview

Hematoporphyrin ether (HPE) is a second-generation photosensitizing agent used in photodynamic therapy (PDT) for the treatment of various malignancies. Developed and manufactured by Shenzhen Zhongxing Yangfan Bioengineering, HPE is a purified fraction of hematoporphyrin derivative (HpD) enriched with ether-linked oligomers, which exhibit superior tumor-localizing properties and photochemical activity. Following intravenous administration, the drug selectively accumulates in tumor cells. Upon irradiation with a 630 nm laser, HPE absorbs light energy and transfers it to molecular oxygen, generating highly reactive singlet oxygen and other reactive oxygen species (ROS). These agents induce oxidative stress, leading to irreversible damage to cellular membranes, mitochondria, and vasculature, ultimately resulting in tumor necrosis and apoptosis. HPE is primarily indicated for esophageal cancer, gastric cancer, and bladder cancer, particularly in cases where surgery is not feasible or as an adjuvant therapy.
