Hemoglobin crosfumaril is a first-generation, human hemoglobin-based oxygen carrier developed as an artificial blood substitute. It consists of human hemoglobin in which the alpha-subunits are intramolecularly cross-linked using diaspirin fumarate, resulting in increased stability and reduced dissociation compared to native hemoglobin. The drug was designed to function as an oxygen carrier for use in situations such as hypovolemia, shock, and vascular disorders where blood transfusion may not be immediately available or feasible. Its mechanism of action is based on mimicking the oxygen-carrying capacity of red blood cells by circulating modified hemoglobin molecules that can transport and deliver oxygen to tissues[1][2][8]. Development was discontinued after clinical trials due to safety concerns and lack of efficacy benefit over standard care[2].
