Hemospan is a novel hemoglobin-based oxygen carrier and plasma expander designed to serve as a blood substitute and to treat conditions such as critical limb ischemia. Developed by Sangart, Hemospan utilizes maleimide poly(ethylene) glycol conjugation to human hemoglobin, resulting in increased molecular size for prolonged intravascular retention and higher oxygen affinity. This design restricts the release of oxygen in resistance vessels (arterioles), thereby reducing vasoconstriction—a common issue with earlier generations of cell-free hemoglobin products. The drug aims to optimize colloidal osmotic pressure and viscosity, enhancing blood flow and tissue perfusion without promoting oxidative reactions or vasoconstriction. Clinical trials have shown that Hemospan is generally well tolerated in humans, with evidence supporting its efficacy in maintaining hemodynamic stability during surgery[1][2][6][8].
