This triple combination therapy consists of henagliflozin (an SGLT2 inhibitor), retagliptin (a DPP-4 inhibitor), and metformin (a biguanide). It is being evaluated by Peking University First Hospital in collaboration with Jiangsu Hengrui Pharmaceuticals for the treatment of newly diagnosed type 2 diabetes mellitus. Henagliflozin works by inhibiting sodium-glucose cotransporter-2 (SGLT2) in the kidneys to reduce glucose reabsorption. Retagliptin inhibits dipeptidyl peptidase-4 (DPP-4), thereby increasing incretin levels to stimulate insulin secretion. Metformin reduces hepatic glucose production and improves insulin sensitivity. The combination is currently being studied in a Phase 4 trial to compare initial triple therapy against a sequential treatment approach.
