Heparin-binding epidermal growth factor (HB-EGF) neutralizing antibodies are a class of monoclonal antibodies designed to target and inhibit HB-EGF, a member of the epidermal growth factor (EGF) family. HB-EGF is synthesized as a membrane-anchored precursor (proHB-EGF) that undergoes proteolytic ectodomain shedding to release a soluble form (sHB-EGF), which binds to and activates the EGF receptor (EGFR/ErbB1) and ErbB4. Overexpression of HB-EGF is associated with tumor progression, metastasis, and chemotherapy resistance in various cancers, including ovarian and colorectal cancers. By binding to HB-EGF, these neutralizing antibodies block its interaction with EGFR and ErbB4, thereby inhibiting downstream oncogenic signaling pathways. Several candidates, such as Kyowa Kirin's KHK2866 and Daiichi Sankyo's U3-1565, have been evaluated in clinical trials for advanced solid tumors.
