Hepatocyte growth-promoting factor (pHGF) is a protein that functions as a potent mitogen for hepatocytes and Kupffer cells, promoting liver cell proliferation and regeneration. It exerts its effects by activating various intracellular signaling pathways, including MAPK, HIF-1, and JAK-STAT cascades, and specifically the JAK2/STAT3/c-MYC pathway, leading to the upregulation of CDK4/6 and acceleration of the G1/S transition in the cell cycle. Beyond its proliferative actions, pHGF also demonstrates significant cytoprotective and anti-apoptotic properties, stabilizing cell membranes, restoring antioxidant enzyme levels (SOD1, SOD2), increasing HO-1 protein, and preventing fibrogenesis by inhibiting the differentiation of hepatic stellate cells. It has been investigated for its potential in treating various liver conditions, including post-hepatectomy liver regeneration, acute and chronic liver injuries, and portal hypertension, as well as for its protective effects in acute lung injury and its role in improving myocardial ischemia in severe coronary artery disease.
