Hepinofovir fumarate (HT-101) is an orally bioavailable small molecule prodrug of tenofovir, developed by Xi'an Xintong Pharmaceutical Research for the treatment of chronic hepatitis B virus (HBV) infection. As a nucleotide reverse transcriptase inhibitor (NRTI), it is intracellularly metabolized into tenofovir diphosphate, which competes with natural deoxyadenosine 5'-triphosphate for incorporation into viral DNA. This incorporation leads to DNA chain termination and the inhibition of HBV DNA polymerase. Hepinofovir is designed to enhance liver targeting and reduce systemic exposure to tenofovir, potentially offering an improved safety profile regarding renal and bone toxicities compared to older tenofovir formulations such as tenofovir disoproxil fumarate (TDF).
