HER-2 pulsed DC1 is an autologous dendritic cell-based cancer vaccine developed primarily by the H. Lee Moffitt Cancer Center & Research Institute, with early contributions from the University of Pennsylvania. The vaccine is produced by isolating a patient's monocytes via leukapheresis, which are then differentiated into type 1 polarized dendritic cells (DC1). These cells are pulsed with HER-2/neu-derived peptides and activated using interferon-gamma (IFN-γ) and a toll-like receptor 4 (TLR4) agonist, such as lipopolysaccharide (LPS). This specific activation protocol is designed to induce high-level production of interleukin-12 (IL-12) and promote a Th1-polarized immune response. The vaccine is primarily investigated for the treatment of HER2-positive breast cancer, including ductal carcinoma in situ (DCIS) and invasive breast cancer, where it aims to stimulate a T-cell mediated anti-tumor response against HER2-expressing malignant cells.
