HER-Vaxx is a B-cell peptide-based vaccine immunotherapy designed to treat tumors that overexpress the human epidermal growth factor receptor 2 (HER2/neu), such as gastric, breast, ovarian, lung, and pancreatic cancers. The vaccine consists of three B-cell epitope peptide sequences derived from the extracellular domain of HER2/neu conjugated to CRM197 with the adjuvant Montanide. It stimulates a polyclonal antibody response against HER2/neu, leading to inhibition of intracellular phosphorylation in the HER2 signaling pathway and mediating antibody-dependent cellular cytotoxicity (ADCC). Clinical studies have shown that adding HER-Vaxx to standard chemotherapy improves overall survival and progression-free survival in patients with advanced or metastatic gastric or gastroesophageal junction adenocarcinoma with no additional toxicity compared to chemotherapy alone. The immune response induced by this vaccine correlates significantly with tumor reduction[1][3][4][5][6].
