HER1-Vax is a therapeutic cancer vaccine designed to induce an active immune response against the Human Epidermal Growth Factor Receptor 1 (HER1), also known as EGFR. Developed by the Center of Molecular Immunology (CIM) in Havana, Cuba, the vaccine consists of the recombinant extracellular domain of the human HER1 protein, typically administered with the adjuvant Very Small Size Proteoliposomes (VSSP) or Montanide ISA 51. Unlike CIMAvax-EGF, which targets the EGF ligand, HER1-Vax directly targets the receptor itself. By stimulating the production of high-affinity polyclonal antibodies against the HER1 extracellular domain, the vaccine facilitates the blocking of ligand-receptor binding and may induce receptor internalization and degradation. This mechanism effectively inhibits the downstream signaling pathways, such as MAPK and PI3K/Akt, that are critical for tumor cell proliferation, survival, and angiogenesis. HER1-Vax is primarily being investigated for the treatment of advanced solid tumors that overexpress EGFR, including non-small cell lung cancer (NSCLC) and head and neck squamous cell carcinoma.
