The HER2×HER2 T cell bispecific antibody is a homodimeric T-cell engaging bispecific antibody (T-BsAb) designed to target HER2-expressing tumor cells. It utilizes a 2+2 IgG-[L]-scFv format, featuring a bivalent IgG backbone for HER2 binding and two anti-CD3 single-chain variable fragments (scFvs) fused to the light chains to recruit and activate polyclonal T cells. This bivalent binding to HER2 is intended to increase avidity and enhance T-cell mediated cytotoxicity against tumors with varying levels of HER2 expression, such as pancreatic ductal adenocarcinoma (PDAC). Research indicates that bivalent binding to the tumor antigen is critical for robust anti-tumor effects in vivo compared to monovalent counterparts, as it significantly improves tumor infiltration by T cells and mediates potent anti-tumor activity in xenograft models.
