**HER2-28ζ CAR-T cells with shJNK** is an experimental, autologous human T-cell therapy engineered with a second-generation HER2-directed chimeric antigen receptor containing a CD28 costimulatory domain and CD3ζ signaling domain, together with a lentivirally encoded short-hairpin RNA that suppresses c-Jun N-terminal kinase 1 and 2 expression. The CAR directs T-cell recognition and killing of HER2-expressing tumor cells, while JNK knockdown modulates NFAT signaling, increases NFATc1-dependent granzyme B expression and degranulation, and enhanced antitumor cytotoxicity in preclinical ovarian cancer models. The construct was reported by researchers at the University of Alabama at Birmingham and remains a preclinical investigational cell-therapy approach for HER2-positive ovarian cancer.
