HER2-affibody NIR conjugate is a near-infrared (NIR) fluorescently labeled affibody conjugate developed for the non-invasive in vivo optical imaging of HER2-expressing tumors. The conjugate consists of a small (~7 kDa) HER2-specific affibody molecule (derived from the Z domain of Staphylococcus aureus protein A) site-specifically conjugated at its C-terminal cysteine residue to a near-infrared fluorescent dye (such as DyLight-750, DyLight-488, or Alexa Fluor 750). It binds with high affinity to the extracellular domain of HER2 without interfering with the binding of therapeutic antibodies like trastuzumab or pertuzumab. This imaging agent is designed to assess HER2 receptor status in breast cancer and other HER2-positive malignancies, potentially facilitating patient selection and monitoring of HER2 expression during therapy.
