Overview

HER2-CAR-T refers to a class of chimeric antigen receptor (CAR) T cell therapies engineered to target the human epidermal growth factor receptor 2 (HER2), which is overexpressed in various solid tumors including breast, gastric, ovarian, colon, bladder, lung, uterine cervix, head and neck, esophageal cancers and sarcomas. These therapies involve collecting T cells from a patient or donor (autologous or allogeneic), genetically modifying them to express a CAR that recognizes HER2 on tumor cells, expanding them ex vivo and infusing them back into the patient. Upon administration, HER2-CAR-T cells bind selectively to HER2-expressing tumor cells and induce cytotoxicity against these cancer cells[1][3][4]. The approach has shown clinical benefit in early-phase trials for high-risk sarcomas and other solid tumors but carries risks such as cytokine release syndrome due to "on-target off-tumor" effects[3][5]. Multiple next-generation designs are under development aiming for improved safety and efficacy[7]. Several companies are developing proprietary versions of HER2-targeted CAR-T products.
