Overview

HER2-CAR T cells are an investigational class of autologous cell therapies where a patient's T cells are genetically engineered to express a chimeric antigen receptor (CAR) targeting the Human Epidermal Growth Factor Receptor 2 (HER2). HER2 is a tyrosine kinase receptor overexpressed in various solid tumors, including sarcomas, glioblastoma, and breast, gastric, and pancreatic cancers. The therapy, developed through collaborations involving Baylor College of Medicine, Texas Children's Hospital, and City of Hope, typically utilizes second-generation CAR constructs with costimulatory domains like CD28 or 4-1BB to enhance T-cell expansion and persistence. Clinical trials, such as the HEROS series, evaluate the safety and efficacy of these cells administered intravenously or peritumorally, often following lymphodepleting chemotherapy and sometimes in combination with immune checkpoint inhibitors to combat the immunosuppressive tumor microenvironment.
