HER2 CAR VLP is an in vivo engineered, lineage-restricted multicellular immunotherapy developed by Ensoma. Utilizing Ensoma's Engenious platform, the therapy consists of a helper-dependent adenoviral virus-like particle (VLP) that targets CD46-positive cells, including long-term hematopoietic stem cells (LT-HSCs) and mature immune cells. The VLP delivers a large genetic payload (up to 35 kb) encoding lineage-restricted anti-HER2 chimeric antigen receptors (CARs) driven by T/NK-cell-specific and myeloid-specific promoters, alongside a hyperactive Sleeping Beauty transposase (SB100X) for stable genomic integration and an MGMTP140K cassette for chemotherapy-based enrichment. Upon in vivo administration, the platform generates a self-renewing reservoir of HER2-targeting CAR-T, CAR-NK, and CAR-macrophage (CAR-M) cells that cooperatively infiltrate, remodel, and control HER2-positive solid tumors.
