HER2 dBiTE is a DNA-encoded bispecific T-cell engager (dBiTE) being developed by Inovio Pharmaceuticals in collaboration with The Wistar Institute. This biologic immunotherapy utilizes Inovio's proprietary DNA-based platform to deliver a genetic sequence encoding a bispecific antibody fragment. Once administered via the CELLECTRA electroporation device, the patient's cells produce the functional bispecific molecule, which simultaneously binds to the Human Epidermal Growth Factor Receptor 2 (HER2) on cancer cells and the CD3 epsilon subunit on T cells. This dual binding facilitates the recruitment and activation of cytotoxic T cells directly to the tumor site. Preclinical studies indicate that this DNA-encoded approach provides a significantly longer therapeutic window—up to four months—compared to traditional protein-based BiTEs, which often require continuous infusion due to short half-lives. The drug is currently in preclinical development for the treatment of HER2-positive malignancies, including advanced breast, ovarian, gastric, esophageal, and endometrioid cancers.
