The HER2 DC1 vaccine is an investigational autologous dendritic cell-based immunotherapy developed by the H. Lee Moffitt Cancer Center and Research Institute for the treatment of HER2-positive breast cancer. The vaccine is produced by collecting a patient's own monocytes via leukapheresis, which are then matured in vitro into type I polarized dendritic cells (DC1) and pulsed with MHC class II-restricted peptides derived from the HER2 (human epidermal growth factor receptor 2) protein. These DC1 cells are specifically designed to secrete high levels of interleukin-12 (IL-12), which promotes the restoration and expansion of anti-HER2 CD4+ Th1 immune responses. Administered via ultrasound-guided intranodal injection, the vaccine aims to stimulate a robust T-cell mediated attack against HER2-expressing tumor cells. It has been evaluated in clinical trials for various stages of the disease, including ductal carcinoma in situ (DCIS) and as a neoadjuvant or adjuvant treatment to prevent recurrence in patients with residual disease.
