HER2 degrading bispecific antibodies are a specialized class of bispecific antibodies engineered to target the Human Epidermal Growth Factor Receptor 2 (HER2) and facilitate its removal from the cell surface via internalization and lysosomal degradation. Unlike traditional HER2-targeted therapies that primarily inhibit signaling or mediate immune-cell killing, these agents often employ a biparatopic binding strategy—engaging two distinct, non-overlapping epitopes on the HER2 protein—to promote extensive receptor cross-linking and clustering. This clustering triggers efficient endocytosis and trafficking to the lysosome for degradation, thereby depleting the total cellular pool of HER2. This mechanism is particularly relevant for treating cancers that have become resistant to standard HER2 inhibitors or those with low to moderate HER2 expression levels.
