The HER2 dendritic cell vaccine is an investigational immunotherapy designed to stimulate the immune system to recognize and attack cancer cells that overexpress the human epidermal growth factor receptor 2 (HER2). The therapy involves extracting a patient's own dendritic cells and exposing them ex vivo to fragments or peptides of the HER2 protein. These "trained" antigen-presenting cells are then reintroduced into the patient—often directly into tumor tissue—to activate T-cell responses specifically against HER2-expressing tumor cells. The mechanism of action centers on enhancing anti-tumor immunity by promoting both antibody-mediated and T-cell–mediated responses targeting the HER2 antigen. Clinical studies have shown promising results in patients with early-stage or locally advanced (stage II/III) breast cancer that is positive for HER2 expression, particularly in estrogen receptor-negative subtypes. The approach has demonstrated safety, immunogenicity, and preliminary clinical benefit as monotherapy or in combination with chemotherapy[6][8][5][3][4].
