Drug intelligence / Profile preview
A HER2-directed cancer vaccine is a type of active immunotherapy designed to stimulate the patient's immune system to recognize and destroy cancer cells that overexpress the human epidermal growth factor receptor 2 (HER2) protein. These vaccines utilize various platforms, including peptide-based (e.g., nelipepimut-S, GP2, AE37), DNA-based (e.g., WOKVAC, HER2 ICD vaccine), or dendritic cell-based (e.g., DC1) technologies. By presenting HER2 antigens to the immune system, the vaccines generate robust, antigen-specific T-cell and antibody responses. This therapeutic approach is primarily investigated for preventing recurrence in high-risk early-stage breast cancer and protecting against the development of brain metastases in advanced HER2-positive breast cancer, as T-cells are capable of crossing the blood-brain barrier.
