HER2-IGFBP2-IGF1R vaccine (WOKVAC) is a multi-antigen, polyepitope plasmid DNA-based therapeutic cancer vaccine developed by the Cancer Vaccine Institute at the University of Washington, in collaboration with EpiThany and Aston Sci. The vaccine consists of the mammalian expression vector pUMVC3 encoding Th1-selective epitopes of three tumor-associated antigens overexpressed in breast cancer: human epidermal growth factor receptor 2 (HER2), insulin-like growth factor-binding protein 2 (IGFBP-2), and insulin-like growth factor 1 receptor (IGF-1R). Administered intradermally in combination with sargramostim (GM-CSF) as an adjuvant, the vaccine transfects local cells to express these epitopes, stimulating a robust and durable type 1 helper T-cell (Th1) immune response against tumor cells expressing these antigens. WOKVAC is being evaluated in Phase I and Phase II clinical trials for the treatment and prevention of recurrence in various breast cancer subtypes, including HER2-positive breast cancer and ductal carcinoma in situ (DCIS).
