Her2-LAMP is an investigational therapeutic DNA plasmid cancer vaccine developed by Immunomic Therapeutics in collaboration with Duke University. Utilizing Immunomic's proprietary UNITE (UNiversal Intracellular Targeted Expression) platform, the vaccine encodes the human epidermal growth factor receptor 2 (HER2/ErbB2) tumor-associated antigen fused in-frame with lysosomal-associated membrane protein 1 (LAMP-1). This fusion directs the expressed HER2 antigen to the endolysosomal compartment of antigen-presenting cells, enhancing antigen processing and presentation via both MHC class II and MHC class I pathways. This mechanism stimulates robust, polyfunctional HER2-specific CD4+ and CD8+ T cell responses as well as humoral antibody responses, promoting tumor regression and infiltration of activated T cells into the tumor microenvironment. It is primarily being investigated for the treatment of HER2-expressing cancers, such as HER2-positive breast cancer.
