HER2-LAMP-sCD40L is a bicistronic DNA vaccine developed by Immunomic Therapeutics using its UNITE (Universal Intracellular Targeted Expression) platform. The vaccine is designed to treat HER2-positive cancers, such as breast cancer, by inducing a robust immune response. It consists of two primary components: the human epidermal growth factor receptor 2 (HER2) tumor antigen fused to lysosomal-associated membrane protein 1 (LAMP-1), and a soluble form of the CD40 ligand (sCD40L) which serves as a molecular adjuvant. The LAMP-1 fusion directs the HER2 antigen to the lysosomal compartment of antigen-presenting cells (APCs) for enhanced MHC class II presentation, while sCD40L activates APCs via the CD40 receptor, promoting T-cell activation and antibody production. Preclinical studies have demonstrated significant tumor growth inhibition and improved survival in HER2-positive tumor models.
