The HER2-mRNA-LNP vaccine is an experimental mRNA-based cancer vaccine encapsulated in lipid nanoparticles (LNPs) designed to target Human Epidermal Growth Factor Receptor 2 (HER2). The vaccine functions by delivering mRNA encoding the HER2 antigen into host cells, which then express the protein to stimulate a systemic, HER2-specific immune response. Preclinical data presented at ASTRO 2025 demonstrated that the vaccine significantly inhibits tumor growth in HER2-overexpressing Lewis Lung Carcinoma (LLC) mouse models. The study also highlighted a synergistic anti-tumor effect when the vaccine is combined with radiotherapy, which appears to enhance the immune response and alter the tumor immune microenvironment. Safety evaluations in animal models indicated no significant toxicity in major organs or blood parameters.
