The HER2-peptide vaccine is a therapeutic vaccine designed to stimulate an immune response against cancer cells that express the Human Epidermal Growth Factor Receptor 2 (HER2) protein. The active peptide component, nelipepimut-S (also known as E75), is derived from the extracellular domain of the HER2 protein. It functions by stimulating CD8-positive cytotoxic T-lymphocytes (CTLs) to recognize and destroy HER2-expressing tumor cells. In clinical trials, this vaccine is often administered in combination with granulocyte-macrophage colony-stimulating factor (GM-CSF), which acts as an adjuvant to enhance the immune response by promoting the proliferation, maturation, and migration of dendritic cells, thereby augmenting antigen presentation. This combination has been investigated for the prevention of recurrence in breast cancer patients, including those with HER2 low-expressing tumors and ductal carcinoma in situ (DCIS).
