The HER2 pulsed dendritic cell vaccine is an investigational autologous cellular immunotherapy designed to stimulate the immune system to recognize and attack cancer cells expressing human epidermal growth factor receptor 2 (HER2). The therapy involves isolating a patient's mononuclear cells via apheresis, differentiating them into type-1 polarized dendritic cells ex vivo, and then "pulsing" these cells with HER2 antigen. The matured, antigen-loaded dendritic cells are then administered back to the patient as a therapeutic cancer vaccine. This approach aims to induce or enhance anti-HER2 T-cell responses in patients with HER2-positive cancers such as metastatic breast cancer or high-risk bladder cancer. In some clinical protocols, this vaccination is combined with other agents (e.g., trastuzumab or pepinemab) and may be followed by adoptive transfer of ex vivo expanded HER2-specific T-cells[3][5][7].
