HER2 siRNA (siHER2) is a small interfering RNA (siRNA) designed to target and silence the expression of human epidermal growth factor receptor 2 (HER2/ErbB2) at the mRNA level. Unlike monoclonal antibodies (such as trastuzumab) or small molecule tyrosine kinase inhibitors (such as lapatinib) which block HER2 protein activity, HER2 siRNA halts the synthesis of the HER2 protein itself. This approach has been shown to overcome both intrinsic and acquired resistance to traditional HER2-targeted therapies in HER2-positive breast cancer and other HER2-overexpressing malignancies. Various delivery systems, including polymer-modified mesoporous silica nanoparticles (such as those developed by PDX Pharmaceuticals), are being investigated to enable effective systemic delivery of HER2 siRNA to solid tumors.
