Overview

HER2 t-haNK is an "off-the-shelf" natural killer (NK) cell therapy that has been genetically engineered to express a chimeric antigen receptor (CAR) targeting **human epidermal growth factor receptor 2 (HER2)**, which is overexpressed in a number of solid tumors, including breast cancer and glioblastoma. The construct uses a HER2-specific scFv antibody fragment for tumor cell recognition and includes a CD28.CD3zeta signaling domain, enhancing NK cell activation. HER2 t-haNK is designed for targeted immunotherapy against HER2-positive cancers, offering precise tumor targeting and high cytotoxicity even against tumor cells typically resistant to NK-mediated killing. Administered as an allogeneic ("off-the-shelf") cell therapy, it is under development for recurrent HER2-positive glioblastoma, with early-phase clinical studies initiated.
