HER2-TAC gamma-delta T cells are an experimental allogeneic cell therapy developed by Triumvira Immunologics. This therapy utilizes the proprietary T cell Antigen Coupler (TAC) technology, which is a multi-domain chimeric receptor designed to co-opt the natural T cell receptor (TCR) complex. The HER2-TAC construct consists of an anti-HER2 antigen-binding domain, a CD3-binding domain that recruits the endogenous TCR, and a CD4 co-receptor domain to facilitate signaling. By engineering gamma-delta (γδ) T cells with this construct, the therapy aims to combine the innate, MHC-independent tumor-killing capabilities of γδ T cells with the high specificity of the TAC platform. Preclinical studies indicate that these cells exhibit potent cytotoxicity against HER2-positive solid tumors, including those resistant to conventional γδ T cell therapies.
