This combination therapy consists of autologous chimeric antigen receptor (CAR) T-cells engineered to target the Human Epidermal Growth Factor Receptor 2 (HER2) and the monoclonal antibody trastuzumab. Developed by the National University Hospital, Singapore, this regimen is being evaluated for the treatment of HER2-positive advanced solid tumors, including breast and gastric cancers. The CAR-T cells are designed to directly recognize and eliminate HER2-expressing tumor cells, while trastuzumab, which also binds to HER2, may enhance the anti-tumor effect through antibody-dependent cellular cytotoxicity (ADCC) and inhibition of HER2 signaling. The treatment protocol typically involves lymphodepleting chemotherapy with fludarabine and cyclophosphamide prior to CAR-T cell infusion.
