Drug intelligence / Profile preview
HER2-targeted monoclonal antibodies are a class of targeted cancer immunotherapies designed to bind specifically to the human epidermal growth factor receptor 2 (HER2), a receptor tyrosine kinase overexpressed in approximately 15-20% of breast cancers as well as other solid tumors like gastric and ovarian cancers. By binding to the extracellular domain of HER2, these antibodies inhibit downstream oncogenic signaling pathways (such as PI3K/Akt and MAPK), prevent receptor dimerization, and induce antibody-dependent cellular cytotoxicity (ADCC) to recruit immune effector cells to the tumor site. Well-known examples of approved HER2-targeted monoclonal antibodies include trastuzumab and pertuzumab, which are widely used as standard-of-care therapies in HER2-positive malignancies.
