HER2-targeting CAR-macrophage cells are an investigational autologous chimeric antigen receptor (CAR) cell therapy developed by The Affiliated Hospital of Xuzhou Medical University. Unlike traditional CAR-T cell therapies, this approach utilizes macrophages as the effector cells, which are genetically engineered to express a CAR specific for the Human Epidermal Growth Factor Receptor 2 (HER2). These CAR-M cells are designed to overcome the challenges of treating solid tumors by effectively penetrating the tumor tissue, initiating direct phagocytosis of HER2-positive tumor cells, and remodeling the immunosuppressive tumor microenvironment into a pro-inflammatory state. This transition facilitates the recruitment and activation of adaptive immune cells, potentially leading to a sustained anti-tumor response. The therapy is currently being evaluated in Phase 1 clinical trials for patients with HER2-positive solid tumors.
