Overview

HER2Bi-armed activated T cells (HER2 BATs) is an autologous cell-based immunotherapy developed by the Barbara Ann Karmanos Cancer Institute and Wayne State University. The therapy involves collecting a patient's T cells via leukapheresis, activating them ex vivo with anti-CD3 monoclonal antibodies, and expanding them in the presence of interleukin-2 (IL-2). These activated T cells (ATC) are then 'armed' with a bispecific antibody (HER2Bi) that targets both the CD3 receptor on the T cells and the HER2 (human epidermal growth factor receptor 2) antigen on tumor cells. This redirection allows the patient's own immune system to target and lyse cancer cells expressing HER2, including those with low levels of expression (HER2 0, 1+, or 2+). The treatment is typically administered as a series of intravenous infusions, often supported by low-dose IL-2 and GM-CSF to enhance T cell persistence and anti-tumor activity.
