Overview

HER2Bi-armed activated T cells (HER2 BATs) is an investigational autologous cell-based immunotherapy developed by the Barbara Ann Karmanos Cancer Institute in collaboration with Wayne State University. The therapy involves extracting a patient's own T cells via leukapheresis, activating them with an anti-CD3 monoclonal antibody, and expanding them in the presence of IL-2 for 14 days. These activated T cells (aATC) are then 'armed' with a bispecific antibody (HER2Bi) that targets both CD3 on the T cells and HER2 (human epidermal growth factor receptor 2) on tumor cells. This dual targeting redirects the patient's immune cells to recognize and attack cancer cells, even those with low HER2 expression (HER2-negative 0, 1+, or 2+). The therapy has been evaluated in Phase I and Phase II clinical trials for metastatic and locally advanced breast cancer, often administered in combination with low-dose IL-2 and GM-CSF to enhance T cell persistence and anti-tumor activity.
