HER3-ADC1 is an antibody-drug conjugate (ADC) designed to target human epidermal growth factor receptor 3 (HER3) expressing cancers. It comprises the human HER3 monoclonal antibody seribantumab, conjugated via a cleavable valine-citrulline linker to the cytotoxic payload monomethyl auristatin E (MMAE) using a stochastic cysteine conjugation method. This ADC has demonstrated target-dependent in vitro cytotoxicity and in vivo anti-tumor activity in HER3-expressing pancreatic cancer patient-derived xenograft models. Developed by Elevation Oncology, HER3-ADC1 aims to selectively deliver a potent chemotherapy agent to cancer cells, thereby minimizing systemic toxicity.
