HER3 dpADC is an unnamed, preclinical **HER3-directed dual-payload antibody-drug conjugate** developed by GeneQuantum for EGFR-mutant non-small cell lung cancer, including osimertinib-resistant disease. The construct couples a HER3 antibody to a novel DNA topoisomerase I inhibitor designated TopoIx and an EGFR tyrosine kinase inhibitor through two orthogonal enzymatic site-specific conjugation approaches and stable linkers. In preclinical cell culture, xenograft, and patient-derived xenograft models, it produced dose-dependent antitumor activity, bystander killing, and tumor regressions without obvious toxicity.
