HER3 monoclonal antibodies are a class of targeted biologics designed to bind to the Human Epidermal Growth Factor Receptor 3 (HER3, also known as ErbB3). HER3 is a member of the receptor tyrosine kinase (RTK) family that is frequently overexpressed in various solid tumors, including non-small cell lung cancer (NSCLC), breast cancer, and gastric cancer. Unlike other members of the EGFR family, HER3 has minimal intrinsic kinase activity and functions primarily by forming heterodimers with other receptors, such as HER2 or EGFR, to activate the PI3K/AKT/mTOR signaling pathway, which promotes cell proliferation and survival. Monoclonal antibodies targeting HER3 work by binding to the extracellular domain of the receptor, thereby blocking the binding of ligands like neuregulin-1 (NRG1) or preventing the receptor from dimerizing with other HER family members. These antibodies are also being extensively developed as the targeting components of antibody-drug conjugates (ADCs) to deliver cytotoxic payloads directly to HER3-expressing malignant cells.
