HER3 novel duocarmycin is a preclinical-stage antibody-drug conjugate (ADC) developed by BioNerviano, a subsidiary of Nerviano Medical Sciences. The therapeutic candidate consists of a monoclonal antibody targeting the Human Epidermal Growth Factor Receptor 3 (HER3/ErbB3) conjugated to a potent duocarmycin-based payload. Duocarmycins are a class of small-molecule toxins that function as DNA alkylators, binding to the minor groove of DNA and causing irreversible alkylation, which leads to DNA damage and apoptosis in target-expressing cancer cells. HER3 is a member of the EGFR family and is frequently overexpressed in various solid tumors, where it plays a key role in tumor growth and resistance to other targeted therapies. This ADC leverages BioNerviano's proprietary conjugation and linker-payload technology to provide a targeted treatment option for HER3-positive malignancies.
