HER3 shRNA is a short hairpin RNA (shRNA) designed to silence the expression of the Human Epidermal Growth Factor Receptor 3 (HER3, also known as ERBB3). By utilizing the RNA interference (RNAi) pathway, the shRNA sequence—often delivered via viral vectors such as pLKO.1—targets HER3 mRNA for degradation, leading to a significant reduction in HER3 protein levels. In preclinical research, particularly in colorectal cancer models, HER3 knockdown has been shown to impede anchorage-independent growth and modulate the HER3-EREG signaling axis, which is associated with disease aggression and poor prognosis. This modality represents a targeted approach to inhibiting HER-family signaling in tumors that may be resistant to traditional EGFR-targeted therapies.
