HER577 is a novel human epidermal growth factor receptor 2 (HER2)-derived HLA-A2-restricted immunogenic peptide vaccine candidate consisting of the nonameric sequence FGPEADQCV (amino acids 577-585) [1271-1271]. Discovered by researchers at the University of Texas MD Anderson Cancer Center, Brooke Army Medical Center, and the Uniformed Services University of the Health Sciences, HER577 is located within the trastuzumab-binding domain of the HER2 protein [1271-1271]. Binding of trastuzumab to HER2 is hypothesized to alter proteasomal cleavage patterns, protecting or modulating enzymatic cleavage sites and thereby enhancing the presentation of HER577 on HLA-A2 molecules [1271-1271]. In preclinical studies, HER577-stimulated cytotoxic T lymphocytes (CTLs) demonstrated specific lysis of HER2-expressing tumor cells, which was significantly enhanced upon pretreatment of the target cells with trastuzumab [1271-1271]. It is being investigated for the treatment of HER2-positive cancers, including breast, ovarian, and prostate cancers, in combination with trastuzumab.
